The role of CXC receptors signaling in early stages of mouse embryonic stem cell differentiation

EBs were also used to analyze the impact of gene overexpression [e.g. (Prelle et al., 2000; Rohwedel et al., 1995)] or treatment with extracellular factors, such as Wnt, on cellular differentiation (ten Berge et al., 2008) on differentiation. Among extracellular factors that can impact ESC differentiation are growth factors and chemokines, i.e. those ones that induce various cellular processes including differentiation or migration.

2011, Life SciencesCitation Excerpt :Total RNA was isolated from whole bladder using the TRI REAGENT™ (Sigma), and RNA was reverse transcribed using the SuperScript™ First-Strand Synthesis System for RT-PCR (Invitrogen). The resulting complementary DNAs (cDNAs) were used for polymerase chain reaction (PCR) with specific primers based on rat cDNA (LaPolla et al., 1984; Buonanno et al., 1989; Witzemann et al., 1990; Rohwedel et al., 1995; Liu et al., 1998; Kullmann et al., 2008a) (Table 1). After an initial denaturation at 94 °C for 2 min, the indicated cycles of amplification [1 min of denaturing at 94 °C, 1 min of annealing at the temperature indicated in Table 1, and 1 min of extension at 72 °C] were performed in a DNA Thermal Cycler (PerkinElmer, Norwalk, CT), iCycler (Bio-Rad, Hercules, CA), and Program Temp Control System PC708 (ASTEC, Fukuoka, Japan).

These include known pluripotency-associated proteins OCT-4, UTF-1, DNMT-3B, CDH-1 (39–47) (embryonic cadherin), and TCF-3 (48). - Fold change also identified 175 proteins enriched after retinoic acid-mediated differentiation in all three experiments (Fig. 3D and supplemental Table S3).

Mesp1 failed to induce other paraxial mesoderm genes, such as Mesp2, Tbx6, or Pax1 (Sakurai et al., 2006) (Figure S3C), or skeletal myogenic transcription factors Myod, Myogenin, or Myf5 (Figures S3A and S3B) (Rohwedel et al., 1995). However, this could either be due to the inability of Mesp1 to induce paraxial genes or to a bias against paraxial gene expression in the culture conditions.
