M-twist Expression Inhibits Mouse Embryonic Stem Cell-Derived Myogenic Differentiation in Vitro

Abstract

The mouse M-twist gene codes for a basic helix-loop-helix protein which was shown to be inhibitory for differentiation of myogenic cells in culture. Mouse embryonic stem (ES) cells of line BLC6 efficiently differentiating into skeletal muscle cells when cultivated as embryo-like aggregates (embryoid bodies) were stably transfected with the plasmid pME18s-twist containing the M-twist gene under the control of the modified SV40 early promoter SRα. Two pME18s-twist-expressing clones showed delayed and reduced skeletal muscle cell differentiation depending on the level of exogenous M-twist expression compared to control cells. By morphological analysis using phase contrast microscopy and hematoxylin-eosin staining, the development of first myocytes and formation of myotubes in embryoid body outgrowths of these clones were found to be delayed for about 3 days in comparison to control cells. Immunofluorescence studies with a monoclonal antibody against sarcomeric myosin heavy chain revealed that myogenic cells appeared in so-called myogenic centers showing a reduced number of myocytes and myotubes in the M-twist-expressing clones. Using RT-PCR analysis the expression of the skeletal muscle determination genes myf5, myogenin, and MyoD as well as muscle-specific genes coding for the γ-subunit of the nicotinic acetylcholine receptor and the cell adhesion molecule M-cadherin were found to appear with a delay of at least 1 to 4 days in the pME18s-twist-transfected cells during the development of embryoid bodies. We conclude that the constitutive expression of the mouse M-twist gene during ES-cell-derived differentiation has an inhibitory effect on skeletal muscle cell development depending on the level of exogenous M-twist expression.

EBs were also used to analyze the impact of gene overexpression [e.g. (Prelle et al., 2000; Rohwedel et al., 1995)] or treatment with extracellular factors, such as Wnt, on cellular differentiation (ten Berge et al., 2008) on differentiation. Among extracellular factors that can impact ESC differentiation are growth factors and chemokines, i.e. those ones that induce various cellular processes including differentiation or migration.

    Total RNA was isolated from whole bladder using the TRI REAGENT™ (Sigma), and RNA was reverse transcribed using the SuperScript™ First-Strand Synthesis System for RT-PCR (Invitrogen). The resulting complementary DNAs (cDNAs) were used for polymerase chain reaction (PCR) with specific primers based on rat cDNA (LaPolla et al., 1984; Buonanno et al., 1989; Witzemann et al., 1990; Rohwedel et al., 1995; Liu et al., 1998; Kullmann et al., 2008a) (Table 1).     Using -fold change alone, we identified 101 proteins >1.5-fold enriched before differentiation in both ES and EC cells in all three data sets (Fig. 3C). These include known pluripotency-associated proteins OCT-4, UTF-1, DNMT-3B, CDH-1 (39–47) (embryonic cadherin), and TCF-3 (48). - Fold change also identified 175 proteins enriched after retinoic acid-mediated differentiation in all three experiments (Fig. 3D and supplemental Table S3).

    Mesp1 induced the paraxial mesoderm genes Pax3 and Tcf15 on day 4, but this was not sustained at day 6 (Figures S3A and S3B). Mesp1 failed to induce other paraxial mesoderm genes, such as Mesp2, Tbx6, or Pax1 (Sakurai et al., 2006) (Figure S3C), or skeletal myogenic transcription factors Myod, Myogenin, or Myf5 (Figures S3A and S3B) (Rohwedel et al., 1995). However, this could either be due to the inability of Mesp1 to induce paraxial genes or to a bias against paraxial gene expression in the culture conditions.
