2021, Molecular CellRegion 4 of the RNA polymerase s subunit counteracts pausing during initial transcription
2021, Journal of Biological ChemistryCitation Excerpt :In the σΔ4.2 fragment (residues 1–553), only region 4.2 was deleted, but not the 4.1 α-helix, which binds inside the RNA exit channel (Fig. 1E). In agreement with previous studies (44, 45), the EcoRNAP holoenzymes harboring the σΔ4 or σΔ4.2 fragments were inactive in abortive transcription assays with the −10/−35 type lacUV5 promoter and displayed reduced transcriptional activity with the “extended −10” type galP1cons promoter (Fig. 1F). The σΔ3.2 subunit, in which residues 513 to 519 in the σ3.2-finger were deleted, was active in transcription initiation with both promoters.
