Position-specific expression of the annulin protein during grasshopper embryogenesis

Abstract

Annulin, named for its annular expression in developing limb buds, is a ∼100 kDa membrane-associated protein that is expressed in a complex and changing pattern during grasshopper embryogenesis. Its expression is dynamic along the developing midline and in the mesoderm, transient in neuroepithelial sheath cells around mitotic neuroblasts, and position-specific in circumferential stripes in each limb bud segment. Annulin expression begins along the midline of the embryo at the onset of gastrulation. Mesoderm cells express the protein as they migrate away from the midline as do new cells that come to lie at the midline. During neurogenesis, annulin expression disappears from many midline cells until only a specific subset of midline glial cells expresses high levels of the protein. Starting at the beginning of neurogenesis, sheath cells express annulin in correlation with the mitotic activity of the neuroblasts they surround.

      The grasshopper antenna is such an appendage, comprising scape, pedicel and flagellum (Chapman, 1982). Externally, the flagellum is characterized by segments called meristal annuli (Chapman, 2002), whose borders are delimited early in embryogenesis by epithelial bands of Annulin (Bastiani et al., 1992), Lazarillo (Boyan and Williams, 2004), and Lachesin (Ehrhardt et al., 2015a) labeling. The early embryonic antenna comprises four such annuli termed A1, A2, A3, A4 starting from the apex.

      Associated with the axonal scaffold established by the pioneers are the axons of sensory cells differentiating in the epithelium (Fig. 1E), and an array of cells called nerve tract associated (NTA) cells which have a different ontogeny. These NTA cells do not express the neuron-specific antigen HRP (Fig. 1D,E; Jan and Jan, 1982), nor such glia-associated antigens as repo (Fig. 4; Halter et al., 1995) and Annulin (Bastiani et al., 1992; Boyan et al., 1995). The NTA cells are first observed prior to the pioneers – at about 30% of embryogenesis.

      Although the functions of laminin and semaphorins have been partly elucidated, the roles of other molecules expressed in the limb bud remain to be determined. Annulin, a transglutaminase, is restricted to epithelial bands in the developing limb bud (Singer et al., 1992; Bastiani et al., 1992; Singer et al., 1995). G-innexin-1, a connexin gap junction protein, is expressed at the trochanter–coxa segment boundary (Ganfornina et al., 1999).
