Publisher Summary

This chapter discusses and evaluates several methods that reduce or avoid the problems presented by whole-cell recording, and are used to prevent rundown in electrophysiological experiments. Cell-attached patch recording usually implies that cytoplasm is maintained on the intracellular surface of the plasma membrane. With this configuration tingle-channel currents are usually observed. However, if channel density is very high or if very large pipettes are used, macroscopic currents may be observed in cell-attached patches. Such currents are modulable by second messengers and do not wash out. These techniques preserve—to varying extents—the integrity of the cytoplasm. Each has its own virtues and pitfalls, and it is unlikely that one method will suffice for all purposes. Usually, the choice of technique involves a compromise between the ability to determine the composition of the intracellular solution and the preservation of the cytoplasm.

  • Modulation of the Ca2+ signaling pathway by celangulin I in the central neurons of Spodoptera exigua

    Citation Excerpt :

Intracellular Ca2 + increases through influx from the external by voltage-gated calcium channel or receptor-operated calcium channel and efflux from the calcium store by RyRs or IP3Rs [23]. The whole-cell (extracellular) patch-clamp technique facilitates the study of currents through single ionic channels from a wide variety of cells [24,25]. To determine whether celangulin I might be involved in calcium signaling transduction, we examined the effects of celangulin I on the central neurons of S. exigua on the calcium homeostasis with the whole-cell patch-clamp and calcium imaging technique.
