The clinical practice data, although showing that FFPE samples do generally yield more DNA mass, also show higher failure (QNS) rates that are reflective of the negative impact of FFPE-induced degradation, effectively reducing the quantity of suitable quality DNA for downstream NGS.28,42–45 Therefore, implemented in the UVMMC clinical NGS workflow for FFPE-derived DNA extracts (surgical and CB) is real-time PCR analysis with nested primers to define the functional length of the genomic DNA fragments.
