A general transcription factor forms a stable complex with RNA polymerase B (II)

Abstract

A general transcription factor (BTF3) has been purified from HeLa whole cell extracts and shown to be required for accurate initiation of transcription from the adenovirus-2 major late promoter (Ad2MLP) and other RNA polymerase class B promoters. We show that purified BTF3 (27 kd) binds to RNA polymerase B (II), forming a complex that is transcriptionally active. We found no evidence that purified BTF3 interacts with DNA or is required for the formation of the stable preinitiation complex.

      Furthermore, this study also suggests that nuclear BTF3 expression could be considered as a prognostic biomarker for HSCC, while a larger patient cohort is needed to evaluate the prognostic value of BTF3 in HSCC. The BTF3, one of the transcription factor class II D (TFIID) related proteins, is necessary to be steadily combined with the transcription factor assembling TATA box at the very early stage of the transcription initiation and forms a stable complex with RNA polymerase II [1,2]. It exists in variety of mammalian cells as an evolutionary highly conserved gene [4].

      The phosphorylation status of Thr-89 will therefore be important for NACA interaction with BTF3. BTF3 is one of several general transcription factors involved in TATA box–mediated proximal promoter transcription via interactions with RNA polymerase II (32, 54). Our own previous data demonstrated that NACA interacts with TATA-binding protein, and stabilizes cJUN heterodimers (9, 11).
