Proteolytic and lipolytic activities of Micrococcus roseus (65), Halomonas elongata (16) and Vibrio sp. (168) isolated from Danish bacon curing brines

Abstract

Viable cells, cell free extracts and extracellular concentrates of Micorcoccus roseus (65), Halomonas elongata (16) and Vibrio sp. (168) isolated from Danish bacon curing brines were examined for lipase, esterase, proteinase and aminopeptidase activities on natural and synthetic substrates. Micrococcus roseus (65) produced one intracellular esterase with affinity for short chain esters, and two intracellular aminopeptidases with affinity for nonpolar amino acids and l-arginine, respectively. One extracellular esteracellular aminopeptidase with affinity for l-proline was also observed. Three intracellular esterases with affinity for short chain esters and a membrane bound esterase with affinity for butyric to capric esters were found in Halomonas elongata (16), but almost no aminopeptidase acitivity was found. Vibrio sp. (168) had four intracellular esterases with affinity for short chain esters and one esterase with affinity for all tested esters. Furthermore, an enzyme, which did not migrate on electrophoretic gels, had activity on all examined esters and tributyrin. Small intracellular activity on l-alanine was observed for this bacterial strain. There was no proteinase activities in the tested bacteria.

      As the dominant bacterium, certain strains of Tetragenococcus may be better adapted to the ham fermentation system's high salt and complex nutrient environment and influence other microorganisms, thereby affecting the composition of the fermentation product. Tetragenococcus is closely related to the quality of fermented foods by its ability to inhibit histamine production and degrade aflatoxin B1 in fermented foods (Li et al., 2018) and produce flavor substances that improve fermented foods (Hinrichsen et al., 1994).       Halomonas sp. is typically found in saline environments. It can produce lipolytic enzymes that promote the degradation of flavor substances and play an important role in the formation of flavor (Hinrichsen, Montel, & Talon, 1994).       Protection of cells, enzymes and DNA by ectoines against the effect of heat, freezing, freeze–thawing, drying, oxidative damage, proteolysis and desiccation has been reported (Ablinger et al., 2012; Andersson et al., 2000; Goller and Galinski, 1999; Kolp et al., 2006; Lentzen and Schwarz, 2006; Lippert and Galinkski, 1992; Malin and Lapidot, 1996; Van-Thuoc et al., 2013). Halomonas elongata, a new species of the family Enterobacteriaceae, was added to the list because of its relevance in meat fermentation (Hinrichsen et al., 1994). As a consequence of the widened scope of the inventory, the genus Zymomonas has been added to the list.
